# Oral Film Specifications and Batch Documentation

How to write a finished-product spec for an oral strip, what a batch record and COA should contain, and what arrives in a lot release packet.

ANSWER IN BRIEF

A specification is a table of attributes, methods, and limits that both parties sign before the lot is made. The batch record proves the lot followed it, the COA reports the results, and the release packet ties film, sachet, carton, and third-party results to one lot number. If any of those four is missing, you do not have a reorderable product.

## A spec is numbers plus methods

"Dissolves fast, tastes good" is a brief. A specification says 22 × 32 mm ± 1 mm; 120 mg ± 10 %; water activity ≤ 0.60; disintegration ≤ 60 s by the named method; melatonin 90 to 110 % of 3 mg by HPLC; content uniformity per USP \<905\>; micro per USP \<2021\> and \<2022\> against \<2023\> limits. Those are example limits, but that is the shape. The buprenorphine/naloxone label shows it in public: each strength's dimensions and drug concentration as % w/w, 22.0 × 12.8 mm at 17.2 % for 8 mg.

## Ingredient files support qualification

For every ingredient: name and form, supplier, lot, supplier COA, allergen and residual-solvent statements where relevant, storage conditions, and the spec you accept it against. Flavor houses ship a confidential formula, so record the flavor code and version. A botanical extract carries a standardization certificate and heavy-metal results that a synthetic vitamin does not.

## The batch record preserves what happened

It joins the approved instructions to what was done: material lots, quantities weighed, mixing order, mass viscosity, coating gap and line speed, dryer set points and web moisture, strip counts, sachet and carton lots, a coding sample, in-process checks, deviations, and the reviewer's signature. A record that cannot say what wet thickness was cast cannot explain a heavy strip later.

## Reconciliation is the honesty check

Every lot balances input against output. For a 15,000-strip pilot an illustrative line reads: 16,400 cut, 480 rejected, 15,920 pouched, 15,000 cartoned into 500 boxes of 30, 90 retained, 30 to ILS Lab, 800 destroyed as line-clearance and coding waste, 0 unaccounted. Small gaps are normal; unexplained gaps are a deviation.

## A COA is not the whole quality plan

The certificate of analysis reports the lot's release results against the spec and nothing else. Our Lot Release Packet adds a certificate of conformance, lot ID, reconciliation, sachet and carton material IDs with artwork versions, and the applicable ILS Lab results. Ask any manufacturer which of those you will receive, and which results are theirs versus a lab's.

## Packaging approvals belong in the file

Controlled dielines, artwork with version numbers, laminate structure in microns, coding format for lot and expiry, count, and case pack. An approved film in an unapproved sachet is an unapproved product. The OTC simethicone strip label is a useful template: dose per strip, the inactive list, "store below 77 °F (25 °C), protect from heat, light and moisture," and count and pouch type per SKU.

## Worked example: what a reorder needs

A hypothetical brand, Marlow Daily, finishes a 500-box pilot of a vitamin D3 and B12 strip. The reorder file holds formula version MD-D3B12-v3 with suppliers; spec rev B; approved dielines with artwork versions; the pilot batch record and reconciliation; the COA, certificate of conformance, and ILS Lab results; the stability protocol with its next pull date; and a change log showing the flavor code moved from v2 to v3 after panel round two. With that file the second lot is a repeat.

## Common questions

**Who is responsible for the specification?**

It is a joint document. The brand is responsible for the label claim and sensory targets; the manufacturer is responsible for the methods and process limits. Both sign each revision.

**How long are batch records kept?**

Agree it in the manufacturing agreement. A common practice is at least one year past the last lot's expiry, and longer if a retailer or export market asks.
